*Abstract
Neutrophils, innate immune cells pivotal for the antibacterial and antiviral defense, fulfill their role through several cellular mechanisms, including phagocytosis. They also form neutrophil extracellular traps (NETs), a reticulated scaffold structure composed primarily of a DNA backbone and several functional proteins. NETs exert their anti-infective activity by trapping invading pathogens and killing them through virulence factor secretion. However, under serious disease conditions such as sepsis or cancer, NETs could trigger adverse reactions in the body, such as inducing tissue damage or providing shelter for dormant cancer metastasis, thereby exacerbating the disease. To reduce NET-related harmful conditions, a promising option is to reduce the number of NETs in the body by injecting DNase-1, which primarily cleaves the NET DNA backbone. However, the direct in vivo DNase-1 injection could provide only a marginal effect because of the short DNase-1 half-life. To overcome this problem, we developed a long-lasting DNase-1 system using medicinal nanomaterials to effectively cleave NETs, thereby reducing NET-related adverse effects and helping to effectively treat various severe diseases.
