Brown adipose tissue (BAT) is critical for increasing energy expenditure, which is vital in addressing obesity and metabolic disorders. In this talk, I will introduce the molecular mechanisms of thermogenesis identified in BAT so far, as well as the therapeutic potential of targeting BAT. Additionally, I will discuss Letm1 domain containing 1 (Letmd1), a novel regulator of BAT function that we have recently identified. Letmd1 emerged as a key gene influencing BAT from our comparative analysis of multiple RNA-seq datasets. We utilized Letmd1 knockout mice to demonstrate its indispensable role in BAT; these mice displayed cold intolerance and exhibited abnormal BAT morphology. Mechanistically, Letmd1 interacts with the chromatin remodeler Brg1/Smarca4, unveiling a potential new pathway that influences the regulation of thermogenic genes.
