Interleukin (IL)-1β plays a crucial protective role against infection; however, their excessive
production can promote the onset of several inflammatory diseases. Therefore,
understanding the regulatory mechanisms of IL-1β secretion is pivotal for treating these
diseases. Inflammasomes are protein complexes that generate active IL-1β. The most
extensively studied inflammasome is the NLRP3 inflammasome, which detects dangerous
extracellular signals and stimulates immune responses.
Mutations in NLRP3 cause the onset of cryopyrin-associated periodic syndrome, whereas
the hyperactivation of the NLRP3 inflammasome is linked to other inflammatory diseases,
including arthritis, diabetes, Crohn’s disease, and asthma, along with certain
neurodegenerative disorders such as Alzheimer’s and Parkinson’s diseases.
Dysregulated NLRP3 activity plays an important role in inflammatory diseases; therefore, it
is clinically essential to identify methods of suppressing the NLRP3 inflammasome.
However, despite their promising efficacy, no Food and Drug Administration (FDA)-approved
NLRP3 inhibitors are currently available due to toxicity concerns.
