Formulation technologies for dry powder Inhaler: A focus on physical surface modification

Inhaled drug delivery via respiratory route has attracted substantial attention in recent decades to achieve a high local concentration for efficacy with reduced systemic exposure and adverse effects or to achieve a rapid absorption for systemic drug delivery and enhancement of bioavailability. In particular, the successful development of pharmaceuticals utilizing dry powder Inhaler hinges on the development of optimized formulations that ensure both physical and chemical stability, while facilitating efficient delivery of the dose to the target respiratory region. This formulation strategy of dry powder Inhaler into a desired quality carries several challenges from both physicochemical and physiological perspectives; 1) design of aerosolized particle with a desired aerodynamic particle size distribution (APSD) for ensuring efficient drug deposition in targeted area of the respiratory tract, 2) physical stability (during storage or aerosolization of particles from the device), 3) overcoming mucus barrier/biofilm and mucociliary clearance. In this presentation, I’d like to provide an introduction on these fundamental concepts in formulation technologies for dry powder Inhaler followed by discussions on several research cases focusing on physical surface modification of inhalable antibiotics powders.
