* 제목 : Exploring RNA-Protein Interactions Through Single-Molecule Full-Length RBP Footprinting

RNA regulation depends on precise interactions between RNA molecules and RNA-binding proteins (RBPs). Understanding these interactions requires detailed, dynamic observations at the single-molecule level. We present a novel full-length RBP footprinting technique that integrates in vivo chemical cross-linking, nanopore direct RNA sequencing, and machine learning algorithms. This approach maps RNA-binding events across entire transcripts, capturing the pseudo-temporal sequence of interactions throughout an RNA molecule’s lifecycle. Our preliminary findings demonstrate this method’s ability to track specific RBP interactions, including the sequential assembly of telomerase complexes and the formation and disassembly dynamics of exon-junction complexes. We are expanding this approach to diverse cellular RNAs to identify how RBP interactions organize into functional modules governing key cellular processes. This comprehensive strategy reveals how RBPs reshape the transcriptome and translatome, providing deeper insights into the molecular mechanisms of gene regulation.
